PeakTrace 1.20 Released

We have released a new version of our PeakTrace DNA base calling software. In addition to the usual bug fixes, this release adds support for the MegaBACE DNA sequencers. At this stage the MegaBACE support is limited to the .abd file type collected with the ET terminator chemistry. This support is still in beta so if you have any problems processing MegaBACE traces please let us know. We will be adding support for the BigDye chemistries in the near future.

How long do you think it will be until you can basecall bigdye megabace traces?

This is hard to say as the reason for the delay is we don't have any example traces from the MegaBACE sequencer that were generated using the BigDye chemistry. Once we have some example traces then it should not take more than a few days. If anyone has such traces then I would be very happy to hear from you.

One big problem of MegaBACE platform is when we have heterozygous deletion mutation template it hard to read the trace because of DYEnamic ET-terminator chemistry have differance molecular weight of each dye-terminator result slip peak overlap (not occur with ABI) that make hard to identify the mutation. Can peakTrace solve this problem?

I don't know as this is not something we have looked at. PeakTrace was not really designed to handle indels. If you want to send us some example traces we could take a look at them as it might do better than the standard MegaBACE base caller.
