We have certainly heard of phred, however, phred is just a basecaller and doesn't offer the same readlengths as either KB or LongTrace. In rather good news we have developed a new basecaller (PeakTrace) that combines the advantages of LongTrace and avoids the need to use KB. We have written PeakTrace so that it can be easily ported to Linux. So far we haven't had any paying demand for a Linux version, but hopefully in the future this will allow us to get away from Windows :)

No there is no linux version of the LongTrace:Box system - I wish there was as I am rather sick of Windows. The reason for this is there is no KB base caller for linux - this means that while we can process the traces on linux, we can't base call them. I don't think many people would like a trace file without a base call!

As for why we used an Apple Mac Mini three reasons: has a powerful CPU, it is small, and silent - the price is pretty good too :)

Some good news for those of you still using the ABI 3700 - we have ben able to create a version of LongTrace that supports traces coming from the ABI 3700. This version is still in testing phase but the results so far suggest that the read lengths obtained are about the same as you can get from ABI 3730 trace (ie in excess of 1000 high quality bases per trace). This translates out at improvements in read length in excess of 40%! If you would like to find out more about this version then please email us.

Using LongTrace:Box to manage demand spikes

Another use of the LongTrace:Box system is to use it to handle sequencing demand spikes. If your demand varies quite a bit you can use LongTrace in two modes. The first mode is when demand is low and you use LongTrace to maximize read length. The second mode is when demand is high and you use LongTrace to increase your sequencer throughput.

This can be a very attractive option if your demand peaks are such that you are thinking of buying a new sequencer - Longtrace is certainly much cheaper than buying a new sequencer!

One of the most exciting options for using LongTrace:Box is to use it to increase the throughput of your ABI DNA sequencer. We have developed a run protocol that will let you do 28 runs per day (~51 min total run time) while keeping the read length of the 12 run per day protocol.

If you are thinking of buying a new sequencer because your demand is growing then the LongTrace:Box might be a much cheaper option - it is certainly much cheaper than a new ABI 3730 sequencer.

The price like everything depends on the volume :) It is in the range of 5 to 10 cents per trace. Given how much it cost to sequence (especially when you include the DNA template prep costs) we think that this is a pretty good deal.
