Hello, pfx taq DNA polymerase by invitrogen recommends you use about 1 unit or approximately 0.5ul of their Pfx in a 50ul reaction. Is there anyone who has used less than what invitrogen recommends? We are tyring to do long PCR reactions with templates with relatively high GC content. Any coomments on volumes lower than 1 unit or 0.5ul for pfx that has given reproduciably good yields with the right prodcut would be of great insight. Thanks
My advice is try using less and see what happens. Also the other thing you can do is reduce the volume of your reaction – I typically do all my PCR reactions at 20µl.
