When doing bidirectional sequencing on a particular exon, the forward sequence is many times very messy (but not always) while the reverse works every time. The sequencing primers are the same as the PCR primers. I haven’t been able to pinpoint the cause, but I believe it is template dependent. Repeating the cycle sequence gives the same result for the same DNA sample. Any thoughts?
